摘要
目的:探讨星形胶质细胞在慢性低O2高CO2大鼠皮层、海马中的表达变化及其与学习记忆的关系。方法:采用慢性低O2高CO2肺动脉高压大鼠模型,30只大鼠随机分为正常对照组(NC组)、低O2高CO22周组(2W组)和低O2高CO24周组(4W组),Morris水迷宫测试评价其学习记忆能力,免疫组织化学法检测皮层、海马胶质纤维酸性蛋白(GFAP)的表达。结果:2W组、4W组大鼠寻找站台的平均逃避潜伏期延长[2W组(28.59±8.35)s,4W组(60.59±17.28)s],游泳总距离增加[2W组(9893.45±1958.16)mm,4W组(18077.57±6878.85)mm],与NC组比较差异具有显著性(P<0.01或P<0.05)。NC组大鼠皮层、海马可见少量GFAP阳性细胞,2W组、4W组较NC组GFAP阳性细胞表达明显增多(2W组皮层41.24±5.76,海马43.37±4.85;4W组皮层50.70±3.38,海马54.67±5.81;P<0.01或P<0.05)。结论:慢性低O2高CO2大鼠空间学习记忆能力下降可能与皮层和海马的星形胶质细胞活化增强有关。
Objective To explore the association of learning-memory with the change of astrocyte in the cortex and hippocampus in rats with chronic hypoxie hypercapnia. Methods Thirty rats were randomized to normal control group (NC group), 2-week hypoxic hypercapnia group (2W group), or 4-week hypoxic hypercapnia group (4W group). The murine model of puhnonary hypertension secondary to chronic hypoxic hypercapnia was then established. Learning-memory was examined in all rats by Morris water maze and the GFAP expression in the cortex and hippocampus was detected by using SP immunohistochemieal technique. Results The mean escape lantency for seeking the platform was prolonged and the overall swimming distance was increased in 2W group [ (28.59 ± 8.35)s and (9 893.45 ± 1 958.16)mm, respectively] and 4W group [(60.59 ± 17.28) s and (18 077.57 ± 6 878.85) ram, respectively], revealing significant differences as compared with NC group (P 〈 0.05 and P 〈 0.01, respectively). The GFAP-positive cells were slightly present in NC group but significantly increased in both 2W (41.24 ± 5.76 in the cortex and 43.37 ± 4.85 in the hippocampus) and 4W group (50.70 ± 3.38 and 54.67 ± 5.81, respectively) (P 〈 0.05 and P 〈 0.01 ). Conclusion The impairment of learningmemory in rats exposed to chronic hypoxic hypereapnia may be associated with the increase of astrocyte activation.
