摘要
目的建立田基黄中槲皮素-7-O-α-L-鼠李糖苷的含量测定方法,为田基黄药材的质量控制和综合开发利用提供一定的科学依据。方法采用HPLC法。色谱柱为Hypersil C18柱(250mm×4.6mm,5μm),流动相为体积分数0.04%的乙腈-磷酸水(体积比77:23),流速为1.0mL·min^-1,检测波长为371nm,柱温为35℃。结果槲皮素-7-O-α-L-鼠李糖苷质量浓度在3.12~99.84mg·L^-1(r=0.9998,n=6)内与峰面积呈良好的线性关系;平均回收率为99.1%(RSD=1.5%,n=9)。结论该方法简便、快速、精密度好,可用于田基黄的质量控制。
Objective To establish an HPLC method for the determination of quercetin-7-O-α-L-rhamnoside in Hypericum japonicum. Methods The separation was performed on an Hypersil C18 column (250 mm×4.6 mm, 5μm) with the mobile phase consisting of 0.04 % acetonitrile-aqueous phosphoric acid ( V : V = 77:23)at the flow rate of 1.0 mL·min^-1. The detection wavelength was set at 371 nm and the column temperature was set at 35 ℃. Results The calibration curve was linear over the range of 3.12 - 99.84 mg·L^-1 for linarin ( r = 0. 999 8). The average recovey was 99.1% (RSD = 1.5 %, n = 9). Conclusions The method is simple, accurate and reproducible, which is suitable for the quality control of Hypericum japonicum.
