摘要
目的探讨丹参对兔耳瘢痕成纤维细胞超微结构的影响,揭示其对瘢痕的作用及机制。方法用丹参对兔耳瘢痕模型进行体内试验,将4只健康的日本大耳白兔的32个耳瘢痕模型随机分为对照组(10个瘢痕)及2个实验组(各11个瘢痕);对照组注射生理盐水,2个实验组分别注射浓度为0.75 g/ml和1.50 g/ml丹参液150 mg和300 mg,于注射后第28、35天分别处死白兔,取材进行HE染色、Masson染色和柠檬酸铅-醋酸铀双染色,采用光镜和透射电镜进行观察。结果对照组成纤维细胞丰富,细胞内粗面内质网发达,未见凋亡小体;2个实验组均表现为瘢痕成纤维细胞减少、胶原合成降低、细胞内粗面内质网减少和凋亡小体产生,而且2个实验组间无明显差别。结论丹参可抑制兔耳瘢痕模型成纤维细胞的生长、增殖及胶原合成。
Objective To explore the effect of salvia miltiorrhiza on the ultrastructure of fibroblasts from rabbit ear scar. Methods A hyperplastic scar model was established using rabbit' ears. Four healthy Japanese big ear rabbits (32 ear scars models) were taken randomly as the control group (10 scars) and two experimental groups (each 11 scars). The control group was injected with normal saline and the experimental groups were injected with 150 mg or 300 mg of 0.75 g/ml and 1.50 g/ml Salvia miltiorrhiza injection respectively. All rabbits were sacrificed at day 28 or 35 after treatment. The pathological changes and ultrastructure of the fibroblasts affecte by light microscope and transmission electron microscope using HE and Masson's stains, and improved lead citrate and urang acetate staining. Results The fibroblasts were numerous, rough endoplasmic reticulum were abundant and no apoptotic bodies were found in the control group. There was no significant difference between the two experimental groups (150 mg and 300 mg of Salvia miltiorrhiza injection). However, compared with the control group, both fibroblasts and rough endoplasmic reticulum of the experimental groups were decreased, there was less collagen synthesis, and apoptotic bodies were found. Conclusion Salvia miltiorrhiza can inhibit the growth, proliferation of fibroblasts and collagen synthesis.
