摘要
通过对铜绿假单胞菌临床分离株的诱变,采用亚胺培南/西司他丁(11,商品名泰能)浓度梯度平皿筛选与液体筛选相结合得到泰能耐药菌株715/R1、715/R2(对泰能的MIC为64μg/ml),连续传2代其耐药性保持稳定;对其测定了对泰能的外膜通透性,并与原泰能敏感菌株作了比较;我们还对它们的膜蛋白进行了SDS—PAGE分析。结果表明:铜绿假单胞菌对泰能的耐药性与外膜蛋白缺失引起的外膜通透率的降低有一定相关性。
Imipenem/cilastatin resistant strains 715/R1 and 715/R2 were obtained by mutagenesis of clinical isolate of Pseudomonas aeruginosa (PA) on this study, their MICs and outer membrane (OM)permeability to imipenem/cilastatin were tested and compared with that of susceptible type strain, their OM proteins were analyzed by SDS-PAGE. The data indicated that PA resistance to imipenem/cilastatin was related to a decrease in OM permeability owing to lacking of a 45 ̄49kD OM protein.
