摘要
目的:研究藤黄酸对人胃癌细胞株BGC-803凋亡的作用及其对胃癌细胞survivin基因表达的影响。观察应用RNA干扰(RNAi)技术沉默survivin基因的表达后,藤黄酸对胃癌细胞BGC-803的抑瘤效应。方法:应用MTT、DNAladder等方法研究藤黄酸对胃癌细胞系BGC-803凋亡的诱导作用,运用RT-PCR、Western-blotting分析藤黄酸和survivin-siRNA对胃癌细胞survivin表达的影响。MTT法检测藤黄酸对转染survivin-siRNA后的BGC-803细胞株抑瘤作用。结果:藤黄酸可抑制BGC-803细胞的生长,其抑制率与药物浓度及作用时间呈依赖关系。藤黄酸作用BGC-803细胞后,survivin蛋白表达降低。通过siRNA有效阻断BGC-803细胞survivin基因的表达后,藤黄酸对细胞的抑瘤效果可以进一步提高。结论:藤黄酸可以有效的抑制survivin基因表达,从而促进肿瘤细胞凋亡,同时survivin-siRNA与藤黄酸具有明显的协同抗肿瘤作用。
Aim: To investigate the effects of gambogic acid induced apoptosis and the influence of gambogic acid on the expression of survivin in human gastric cancer cell line BGC-803, and to explore the inhibitory effects of gambogic acid in human gastric cancer cell line BGC-803 which was subjected to RNA interference (RNAi) to knockdown the expression of survivin gene. Methods: Methyl thiazolyl tetrazolium (MTT) and DNA ladder were used to study the effects of gambogic acid-induced apoptosis in gastric cancer cell line BGC-803. The influence of gambogic acid on the expression of survivin gene in gastric cancer cells was evaluated by RT-PCR and Western blotting, and the chemosensitivities of transfected cells were determined by MTT. Results: Gambogic acid was found not only to down-regulate the expression of survivin but also to inhibit the growth of BGC-803. Induction to the cell apoptosis occurred in dose and time dependent manners. The level of survivin expression was also effectively suppressed by RNAi in a dose-dependent manner. BGC-803 cells of silenced survivin were more sensitive to the treatment of gambogic acid. Conclusion: Gambogic acid could effectively inhibit survivin gene expression and promote tumor cell apoptosis. Sequence specific siRNA targeting survivin could effectively suppress the expression of surviving gene and significantly enhance the chemosensitivity to gambogic acid in gastric cancer cell line BGC-803.
