摘要
目的:探讨不同类型脑星形细胞瘤组织中基质金属蛋白酶-9(MMP-9)的表达与微血管密度(MVD)的关系及其意义。方法:采用免疫组织化学(EnVision)方法检测50例星形胶质细胞瘤组织中MMP-9和CD34的表达,并通过CD34免疫组化结果测定肿瘤组织中MVD。结果:MMP-9在各型脑星形细胞瘤中均有不同程度的表达;不同组织学类型其阳性表达的强度存在统计学差异(Р<0.01)。Ⅲ~Ⅳ级脑星形细胞瘤组织中MMP-9的阳性表达为(76.52±3.38)%,MVD为(87.00±4.18)/高倍视野(HPF);Ⅰ~Ⅱ级中MMP-9的阳性表达为(27.90±2.53)%,MVD为(48.62±4.05)/HPF。Ⅲ~Ⅳ级中MMP-9和MVD的阳性表达明显高于Ⅰ~Ⅱ级(Р<0.01);脑星形细胞瘤中MVD和MMP-9的表达呈正相关。结论:MMP-9的表达和MVD与脑星形细胞瘤的组织学类型及恶性程度相关,有可能做为判断肿瘤恶性程度及预后的辅助指标。
Objective:To investigate the expression of MMP-9 in different kinds of astrocytoma,and their correlation with microvessel density(MVD),and relation with malignancy degree of astrocytoma.Methods:Immmunohistochemical staining was performed to detect the expression of MMP-9 and CD34 in astrocytoma of 50 cases.Expression of CD34 was used to estimate MVD.Results:Expression of MMP-9 protain were detected in all cases in astrocytoma.Positive rates were different in various astrocytomas.There are significant difference between the various groups(P〈0.01).Positive rates of MMP-9 was(76.52±3.38)% and MVD was(87.00±4.18)/HPF in WHO Ⅲ~Ⅳ grade astrocytomas.In WHO Ⅰ~Ⅱ grade,the expression levels were(27.90±2.53)% and MVD was(48.62±4.05)/HPF.The positive rates of expression of MMP-9 in WHO Ⅲ~Ⅳ grade was higher than in WHO Ⅰ~Ⅱ grade(P〈0.01);The expressions of MVD,MMP-9 present positive correlation in tissues of astrocytoma.Conclusion:It suggested that expression levels of MMP-9 and MVD were associaed with histological types and malignancy degree and may be markers to judge malignancy degree and prognosis in astrocytomas.
