摘要
目的:分离大鼠胚胎的脊髓神经干细胞进行体外培养,探讨骨骼肌植块诱导其向运动神经元样细胞分化。方法:应用无血清培养技术分离培养脊髓神经干细胞,通过5-溴-2-脱氧尿苷标记、免疫荧光染色检测细胞增殖、分化情况;通过与骨骼肌植块共培养观察神经干细胞向运动神经元样细胞的分化情况。结果:大鼠胚胎脊髓可成功分离神经干细胞,分化后可表达神经元、星形胶质细胞的特异性抗原;诱导分化后结果显示,骨骼肌植块组有7.87%的细胞呈胆碱乙酰基转移酶阳性,与10%胎牛血清组(0.94%)比较具有统计学意义。结论:从大鼠胚胎脊髓可成功分离神经干细胞,骨骼肌植块可诱导脊髓神经干细胞分化为运动神经元样细胞。
Objective: To isolate and culture neural stem cells from rat embroyonic spinal cord and induce neural stem cells to differentiate into motor neurons like cells when skeletal muscle tissues were used. Methods: Neural stem cells from the rat spinal cord were isolated and cultured in serum free medium. The neural stem cells were labeled by 5-bromo-2-deoxyuridine (BrdU), the nestin and neural cell specific antigens were detected using single or double immunofluorescence staining. Skeletal muscle tissues were used to induce the neural stem cells to differentiate into motor neurons like cells. Results: Neural stem cells could be isolated from the rat embryonic (Edl 4) spinal cord successfully and strongly expressed nestin antigen. They could differentiate into neurons and astrocytes. The statistical results showed that the cell density of ChAT positive cells was 7.87% and significantly higher when exposed to skeletal muscle tissue, compared with 0. 94% in 10% fetal bovine serum group. Conclusion: Neural stem cells may be isolated from rat embryonic spinal cord successfully and skeletal muscle tissue may induce neural stem cells to differentiat into motor neurons like cells.
