摘要
目的通过观察FasL在肺泡巨噬细胞(AM)中的表达,探讨氯化钆(GdCl3)对重症急性胰腺炎(SAP)大鼠所致肺损伤的防护机制。方法将48只成年SD大鼠随机分为三组各16只。对照组不予任何处理。模型组和治疗组逆行性胰胆管注射5%牛磺酸钠建立SAP大鼠模型,治疗组造模后即刻由阴茎背静脉注入2%GdCl315 mg/kg。6 h后处死大鼠,经支气管肺泡灌洗液(BALF)获取肺泡AM,检测BALF中TNFα、IL-1β水平及肺组织髓过氧化物酶(MPO)活性,肺、胰腺组织行HE染色检查,透射电镜观察和流式细胞仪检测肺泡AM凋亡情况,同时行免疫组化法检测肺泡AM的FasL表达。结果与对照组、模型组比较,治疗组透射电镜可见肺泡AM典型凋亡形态学特征,肺组织中MPO活性、BALF中TNFα和IL-1β水平均较模型组降低,肺泡AM凋亡率明显增高,FasL阳性表达率亦明显增高(P均<0.05)。结论GdCl3对SAP所致肺损伤具有防护作用,其机制可能通过激活FasL通路诱导SAP大鼠肺泡AM发生凋亡发挥作用。
Objective To discuss the protection mechanism of gadolinium chloride on lung injury in severe acute panereatitis rats by observing FasL expression in alveolar maerophages. Methods 48 adult experimental SD rats were randomly divided into three groups( 16 in each group) : the normal control group, the severe acute pancreatitis group and the gadolinium chloride treatment group. The ra: model with severe acute pancreatitis was induced by retrograde common bile duet injection of 5% sodium taurocholate. Alveolar maerophages were obtained by bronehoalveolar lavage fluid 6 hours after the model made. The level of TNF-α and IL-1β and activity of MPO were observed. The lung tissue and pancreas tissue were observed by histology. The morphological changes of alveolar maerophages'apoptosis were observed by transmission eleetrie microscopy. The expression of FasL in alveolar maerophages was analyzed by immunocytochemistry. Results The typical morphologic feature of apoptosia could be observed in alveolar macrophages of the gadolinium chloride treatment group. Moreover, the level of MPO and TNF-α and IL-1β decreased significantly compared with the severe acute panereatitis and the normal control group ( P 〈 0.05 ), while the rates of apoptosis of alveolar maerophages and positive rate FasL increased significantly ( P 〈 0. 05 ). Conclusions Gadolinium chloride may induce apoptosis of alveolar maerophages by activating FasL pathway, thus ameliorating lung injury associated with severe acute panereatitis in rats.
