摘要
通过L9(34)正交试验设计,以粉碎粒度60目、萃取时间10min、萃取温度100℃、固液比1/20为最优微波萃取条件,提取盾叶薯蓣中的皂苷元,得率为1.837%,纯度为57.84%,明显优于目前常规的酸水解法和预发酵法。薯蓣皂苷元粗品过酸性氧化铝柱,经苯、石油醚、乙酸乙酯梯度洗脱,采用HPLC检测,纯度达到84.31%,得率为1.21%。最后经红外光谱鉴定,所得产物与标准品结构一致,表明微波提取薯蓣皂苷元结构稳定,提取简便、快速、得率高。
Through analysis of L9(3^4) orthogonal experimental, the results showed that the optimal conditions of microwave extraction: crush size was 60, extraction time was 10 min, extraction temperature was 100 ℃, solid-liquid ratio was 1/20, diosgenin the extraction rate was 1,837 % and purity was 57.84 %. It was superior to the current Standing Orders Acid Hydrolysis solution and pre-fermentation. Using acidic alumina eluting crude as benzene, petroleum ether and ethyl acetate of the diosgenin, the purity reached to 84.31 %, rate was 1.21% by HPLC detection. Finally, from the product structure in line with standard by IR, the microwave extraction of diosgenin structural was stability, simple, rapid and high yield.
