寿胎丸对自然流产模型小鼠母胎界面TNF-αmRNA和IL-4mRNA表达的影响 被引量:5

摘要 目的:探讨寿胎丸对反复自然流产模型小鼠蜕膜组织Th1/Th2型细胞因子TNF-αmRNA和IL-4mRNA表达的影响。方法:设立正常妊娠与自然流产模型,并将自然流产模型小鼠随机分为4组,分别为模型组,寿胎丸低剂量组,寿胎丸中剂量组,寿胎丸高剂量组,应用原位杂交法检测孕14 d各组蜕膜组织TNF-αmRNA和IL-4mRNA的表达。结果:TNF-αmRNA和IL-4mRNA在各组均有表达,与正常妊娠组相比模型组TNF-αmRNA明显升高(P<0.01),IL-4mRNA表达明显降低(P<0.05);与模型组相比寿胎丸低、中、高剂量组TNF-αmRNA表达明显降低(P<0.01),IL-4mRNA表达明显升高(P<0.05),但与正常妊娠组相比均无明显差异(P>0.05)。结论:寿胎丸可能通过降低自然流产小鼠模型TNF-αmRNA表达、升高IL-4mRNA表达,从而调控Th1/Th2型细胞因子的平衡,以达到治疗反复自然流产的目的。 Objective:To study the effect Shou Tai Wan makes on the expression of Th1/Th2 cytokines:TNF-αmRNA and IL-4mRNA at the decidua tissues of mice with recurrent spontaneous abortion.Mothods:Build mice models with normal pregnancy and with spontaneous abortion,divid the mice with spontaneous abortion into 4 groups randomly:model group, Shou Tai Wan low dose group, Shou Tai Wan middle dose group , Shou Tai Wan high dose group, detect the expressions of TNF-αmRNA and IL- 4mRNA at the decidua tissues perganting for 14 days of each group by in situ hybridization.Results:TNF-αmRNA and IL-4mRNA all has expressions in each group, compared with the group with normol pregnancy, the expression of TNF-αmRNA obviously increased in model group(P〈0.05),the expression of IL-4mRNA obviously decreased(P〈0.05); compared with the model group, in Shou Tai Wan low,middle and high dose group ,the expression of TNF-αmRNA obviously decreased(P〈0.05), and the expression of IL-4mRNA obviously increased(P〈0.05),but compared with the group with normol pregnancy,there was no obvious differences(P〈0.05).Conclusion:Shou Tai Wan may regulate the blance of Th1/Th2 cytokines to attain the purpose of curing recurrent spontaneous abortion,by decreasing the expression of TNF-αmRNA and increasing the expression of IL-4mRNA on the mice models with spontaneous abortion.
