摘要
目的探讨热休克蛋白70(HSP70)体外诱导人外周血单个核细胞(PBMCs)后CC型趋化因子CCL5/RANTES在Vγ9Vδ2 T细胞表达。方法人PBMCs在体外用重组HSP70诱导活化,在第1、4、7、10、14天分别收集细胞,流式细胞术胞内细胞因子分析技术检测表达CCL5/RANTES的Vγ9Vδ2 T细胞百分率,并设结核杆菌低相对分子质量抗原(Mtb-Ag)刺激组作为对照。结果HSP70诱导组分泌CCL5/RANTES的Vγ9Vδ2 T细胞百分率在第1、4、7、10、14天分别为5.9±1.7、16.2±2.5、22.8±3.7、32.4±3.6与57.8±7.2;Mtb-Ag诱导组分别为3.7±2.5、10.5±1.8、11.1±2.3、26.3±3.9与47±4.7;HSP诱导Vγ9Vδ2T分泌CCL5/RANTES的能力比Mtb-Ag强。结论HSP70特异性诱导Vγ9Vδ2 T细胞增殖并分泌CCL5/RANTES,表明二者协同作用,共同参与免疫应答并可能介导某些炎症反应。
Objective To explore the secretion of CC-type chemokine CCL5/RANTES from Vγ9Vδ2 T cells in PBMCs stimulated with HSP70 in vitro. Methods Human PBMCs were stimulated with recombinant HSP70 in vitro, and then harvested on day 1, 4, 7, 10, and 14, respectively. The FCM intracellular cytokine analysis was used to detect the percentages of CCL5/RANTES-secreting Vγ9Vδ2 T cells to total homogeneous group, with a control of a low molecular peptide antigen extracted from Mycobacterial tuberculosis. Results The percentages of CCL5/RANTES-secreting Vγ9Vδ2 T cells induced by HSP70 were 5.9 ± 1.7, 16.2 ± 2.5, 22.8 ± 3.7, 32.4 ± 3.6, and 57.8 ± 7.2, respectively; while the data in Mtb-Ag-stimulated gYoup were 3.7 ± 2.5, 10.5 ± 1.8, 11. 1 ± 2.3,26.3 ± 3.9 and 47 ± 4.7, respectively. HSP70 was more effective in inducing Vγ9Vδ2 T cells to secret CCL5/RANTES. Conclusion HSP70 can specifically induce Vγ9Vδ2 T cells to proliferate and secret CCL5/RANTES, which indicates the two effects act synergetically in immune response and may play an important role in some inflammations.
