重组人瘦素蛋白对甲状腺相关眼病眼眶前脂肪细胞增殖和移行的影响 被引量:2

Effects of rh-leptin on the Proliferation and Migration of Orbital Preadipocytes of Thyroid Associated Ophthalmopathy

摘要 目的探讨重组人瘦素蛋白(rh-leptin)对体外培养的甲状腺相关眼病(TAO)患者眼眶前脂肪细胞增殖、移行的影响。方法体外培养TAO眼眶前脂肪细胞,鉴定并传代,用10、50、100、500 mg/L质量浓度的rh-leptin处理,MTT法检测细胞增殖情况,划线法检测细胞移行情况,与空白对照组(0 mg/L rh-leptin)比较。结果500 mg/L的rh-leptin能明显促进眼眶前脂肪细胞的增殖,与对照组相比,差异有统计学意义(P<0.05),但10~100 mg/L组未能观察到具有统计学意义的促增殖效果;10~500 mg/L的rh-leptin能促进眼眶前脂肪细胞的移行,其中50~500 mg/L组与对照组相比,差异有统计学意义(P<0.05)。结论rh-leptin能够促进眼眶前脂肪细胞的增殖、移行,提示瘦素可能参与了TAO的发病机制。 Objective To investigate the effects of rh-leptin on the proliferation, migration of orbital preadipocytes of thyroid associated ophthalmopathy(TAO). Methods Preadipocytes were cultured from orbital tissues of TAO patients after orbital decompression. The subcultured preadipoeytes were incubated with different concentrations of rh-leptin. The proliferation of preadipocytes was tested by methyl thiazlyl tetrazolium (MTT) assay, and the migration was evaluated by crutch method. Results With the treatment of 500 mg/L rh-leptin, the preadipoeytes proliferated more than control(P〈0.05), while 10-100 mg/L rh-leptin has no such significant effects. The migration of the preadipocytes was enhanced by rh-leptin, which showed statistic difference in 50-500 mg/L rh-leptin groups when compared with control group (P〈0. 05). Conclusion Rh-leptin could improve the proliferation and migration of TAO orbital preadipoeytes in vitro, which suggests that leptin may play an important role in the pathogenesis of TAO.
