摘要
目的探讨幽门螺杆菌空胞毒素(VacA)与血小板的相互作用。方法采用流式细胞仪检测VacA与血小板的结合;蔗糖梯度离心法分离血小板细胞膜脂质筏(GEMs),用Western blot检测VacA在血小板表面的结合部位;生物素标记血小板后,用免疫沉降法检测与VacA结合的血小板表面蛋白。结果VacA结合于血小板表面,并存在于GEMs上,结合蛋白的分子量为140kd和40kd。结论VacA可能通过与血小板表面的特异蛋白结合于GEMs上而参与血小板的活化过程。
Objective To investigate the binding reaction of VacA to platelets. Methods Binding of VacA to platelets was detected by flow cytometry. Glyeosphingolipid-enriched microdomains(GEMs) of platelets were isolated by gradient centrifugation and western blot was utilized to demonstrate the position located on the surface of platelets where VacA was bound. The proteins binding with VacA on platelet's surface was detected by immunoprecipitation. Results VacA was shown to be able to bind to platelets and the binding place was further localized in GEMs. Two kinds of binding protein on the surface of platelets were detected by immunoprecipitation, which were 140 kd and 40 kd respectively. Conclusion VacA could bind to specific proteins of GEMs on the surface of platelets, through which platelets were activated.
