摘要
[目的]建立TaqMan-实时荧光PCR快速检测副溶血性弧菌(Vibrio parahemolyticus,VP)的方法,用于食物中毒快速诊断和食品中VP的污染状况调查。[方法]根据GenBank公布的VP tdh基因序列,设计引物和探针,建立TaqMan-实时荧光PCR快速检测体系,应用于食品中VP检测。[结果]TaqMan-实时荧光PCR快速检测体系灵敏度为1.3×104cfu/ml(33 cfu/PCR反应体系),特异性好,无交叉反应。[结论]TaqMan-实时荧光PCR检测体系灵敏度高,特异性强,能提高VP的检出率和准确性,可应用于VP食品污染状况调查和食物中毒的快速诊断。
[ Objective] To establish a TaqMan real-time PCR for rapid detection of Vibrio parahemolyticus (VP) in food poisoning and food contamination. [Methods] A set of primers was designed based on the sequence of tdh gene published in GenBank to detect Vibrio parahemolyticus. The corresponding TaqMan probe labeled with FAM was designed. The TaqMan real-time PCR assay was applied to detect Vibrio parahemolytieus in food. [Results] The sensitivity of the established method was 1.3×10^4cfu/ml (33cfu/PCR reaction) . There was no cross-reaction with other tested bacteria. [Conclusion] The established TaqMan real-time PCR assay is rapid, sensitive and specific. It could be applied to the rapid diagnosis of Vibrio parahemolyticus food poisoning and investigation of the contamination status of it in food.
