摘要
采用单因子和正交试验法,对链霉菌NK413的产抗菌物质的培养基配方和培养条件进行了优化。结果表明,采用甘油10g·L-1,黄豆饼粉20g·L-1,K2HPO40.75g·L-1,MgSO40.25g·L-1,CaCO32g·L-1,NaCl2g·L-1的培养基配方,250mL三角瓶装液量30mL,25℃,180r·min-1离心培养96h,抑菌圈直径可达39mm。应用该优化条件进行70升罐规模发酵中试,证实该发酵条件适合生产应用。
Culture medium and fermentation conditions for the antibiotics produced by Streptomyces NK413 were optimized with single factor and orthogonal design experiment. The best medium recipe was with 10 g·L^-1 glycerol, 20g·L^-1 soybean cake powder, K2HPO4 0.75 g·L^-1,MgSO4 0.25 g·L^-1,CaCO3 2 g·L^-1,NaCl 2 g·L^-1.The optimal fermentation conditions were as follows: 30 mL of medium in 250 mL Erlenmeyer flask with 8 % inoculum under orbital shaking at 180 r/min for 96 h at 25 ℃. Under these conditions, the diameter of inhibition zone of antibiotic produced by strain NK413 was up to 39mm. The preliminary experiment was carried out in 70 L fermentor according above-mentioned culture and conditions, and result indicated that this optimal fermentation conditions could be used to manufacture.
