摘要
目的建立以高效毛细管电泳法测定苯巴比妥片中苯巴比妥含量的方法。方法采用熔融石英毛细管柱(37cm×50μm)作为分离通道;以含75mmol·L^-1十二烷基硫酸钠(SDS)的30mmol·L^-1磷酸氢二钠(pH=8.0)的溶液为缓冲液;分离电压18kV;分离温度30℃,紫外检测波长280am。结果苯巴比妥在8.75—200.00μg·mL^-1浓度范围内线性关系良好(r=0.9979),平均回收率为98.47%,日内和日间RSD均〈5%。结论该方法操作简便、快速、进样量少,成本低,结果准确,可用于苯巴比妥片中主药的含量测定。
Objective To establish a HPCE method to determine the concentration of phenobarbital in phenobarbital tablets. Methods The uncoated fused-silica capillary(30 cm × 50 μm) was used. The eleetrophoresis condition followed as: running voltage was 18 kV, separation temperature was 30 ℃, UV detection wavelength was 280 nm and the running buffer was 30 mmol · L^-1 Na2 HPO4 ( pH = 8.0) containing 75 mmol · L^-1 SDS. The sampler was injected by pressure for 10 s. Results A good linear relation ( r = 0. 997 9 ) was observed in 8.75 - 200.00 μg · mL^-1 concentration of phenobarbital . The average recovery rate of phenobarbital was 98.47%. The RSD of intra-day and inter-day were all less than 5%. Conclusion The established method was simple and fast, and able to control the quality of phenobarbital tablets.
