摘要
研究了二氨基蓝3R(Diamine blue 3R,DR)与牛血清蛋白(BSA)作用的共振光散射(RLS)光谱特征。考察了各种影响因素,并计算出了BSA与DR的结合比(质量之比)为7.37。结果表明,在优化条件下体系的RLS强度与蛋白质浓度在一定范围内具有良好的线性关系,据此建立了一种蛋白质测定的新方法。在最佳实验条件(DR为1.6×10-5mol/L,cHCl=4.0×10-2mol/L)下,BSA的线性范围是0.01~4.5μg/mL,检出限为1.25ng/mL。该方法已成功用于合成样品及尿液的测定。
The determination of proteins with diamine blue 3R by resonance light scattering (RIS) technique was studied. At cHCI = 4.0 × 10^-2 mol/L,a new method for the determination of proteins based on the enhancement of RLS was developed. The linear ranges of the calibration graphs were 0.01 - 4.5 μg/mL for bovine serum albumin (BSA). The corresponding determination limit was 1.25 ng/mL. Samples with foreign substances and urine samples were analyzed satisfactorily.
