摘要
目的:探讨培养液中添加17β-雌二醇(17β-E2)对小鼠生殖泡期(GV)带卵丘细胞卵母细胞体外成熟、受精及其胚胎发育的影响。方法:小鼠生殖泡期卵母细胞随机分为2组:对照组(n=61,TCM199基础培养液加体积分数10%胎牛血清加75 U/L卵泡刺激素加0.5 kU/L人绒毛膜促性腺激素加0.05 g/L青霉素加0.075 g/L链霉素)和17β-E2组(n=66,对照组液加1 mg/L17β-E2),培养后对成熟的卵母细胞行胞浆内单精子显微注射,观察受精及胚胎发育情况。结果:17β-E2组卵母细胞的桑葚胚、囊胚形成率均高于对照组(P均<0.05)。结论:成熟培养液中加入17β-E2有助于小鼠未成熟有卵丘细胞卵母细胞的体外成熟及受精后胚胎发育。
Aim: To study the effects of 17 beta-estradiol(17β-E2) in culture medium on in vitro maturation,fertilization,and embryo developmental competence of germinal vesicle(GV) stage cumulus-oocyte complex(COC).Methods:GV stage oocytes were arranged randomly into two groups: control group(TCM199 plus 10% FBS,75 U/L FSH,0.5 kU/L hCG,0.05 g/L penicillin G,0.075 g/L streptomycin sulfate) and 17β-E2 group(control group medium plus 1 mg/L 17β-E2).After being cultured,mature oocytes were inseminated by intracytoplasmic sperm injection.The fertilization and embryo development were observed.Results: Morula rate and blastocyst rate of COC in 17β-E2 group increased significantly as compared with control group(P〈0.05).Conclusion: Adding 17 beta-estradiol to culture media contributes to maturation in vitro and embryo development after insemination of immature mouse oocytes.
