犬瘟热病毒基因免疫的探索 被引量:2

摘要 将15只经检测未感染犬瘟热病毒(CDV)且抗体阴性的3月龄普通犬随机分为2组,其中试验组10只,对照组5只。将CDV附着蛋白(H)重组质粒pcDNA-H和融合蛋白(F)重组质粒pcDNA-F混合后,以聚乙烯胺(PEI)作为免疫佐剂(各质粒终浓度均为200μg.mL-1),肌肉接种试验组动物;并以生理盐水接种对照组。病毒中和试验表明:动物在首次免疫后抗CDV抗体中和价为10-0.78±0.14,第2次免疫后为10-1.12±0.20,第3次免疫后达到10-1.55±0.19。在试验组第3次免疫后28 d,所有动物均接种CDV分离株YZ0101;结果发现试验组动物均未出现典型的犬瘟热临床症状,且攻毒后第18天外周血淋巴细胞CDV检测为阴性,而对照组动物均出现典型的犬瘟热临床症状和组织病理变化,且攻毒后第18天外周血淋巴细胞中病毒RNA检测为阳性;这表明用所制备的重组质粒免疫动物对抵抗CDV感染具有保护作用。 Total 15 healthy three-month-old dogs were assigned to two groups,while 10 in the first group and 5 in the second group.The first group was immunized with the recombinant plasmids pcDNA-H and pcDNA-H(each 200 μg·mL^-1) which contain the haemagglutinin(H) protein and the fusion(F) protein genes of Canine Distemper Virus(CDV) respectively.To increase the immune response,Polyetherimide(PEI) was used as adjuvant.The second group was only injected with normal saline water as control.The data of virusneutralization test showed that the antibody specific to CDV were 10^-0.78±0.14,10^-1.12±0.20 and 10^-1.55±0.19 after the 1^st,2^nd and 3^rd immunization respectively.All the animals were inoculated with the CDV isolate YZ0101 after the 3^rd immunization,and the data showed that no Canine Distemper clinic symptom and no CDV in the peripheral lymphocyte in the first group,while all the 5 animals in the control group were infected with CDV.This study revealed that the recombinant plasmids pcDNA-H and pcDNA-H could be used as CDV gene vaccine.
