姜黄素抗As_2O_3诱变作用的实验研究

摘要 目的:研究姜黄素抗三氧化二砷(As2O3)的诱变作用。方法:分别采用小鼠骨髓细胞和人外周血淋巴细胞微核(MN)实验,计数含有MN的嗜多染红细胞和淋巴细胞数,计算MN率。观察As2O3细胞诱变性及不同浓度姜黄素对As2O3诱变性作用的影响。结果:①小鼠骨髓MN试验:与As2O3组比较,As2O3+1.2mg/mL,2.4mg/mL,4.8mg/mL姜黄素组小鼠骨髓细胞MN率显著降低(P<0.05,P<0.01)。②人外周血淋巴细胞MN试验:与空白对照组比较,1μg/mL、2μg/mL、4mg/mLAs2O3组人外周血淋巴细胞MN率升高(P<0.01);与1μg/mL(2μg/mL、4μg/mL)As2O3组比较,5μg/mL,10μg/mL,20μg/mL姜黄素+1μg/mL(2μg/mL,4μg/mL)As2O3组MN率降低(P<0.05,P<0.01)。结论:As2O3具有剂量相关的致突变作用,姜黄素具有明确的抗As2O3诱变性作用。 Objective :To observe the inhibitive effect of curcumine against mutagenicity of As2O3. Methods: (1)Micronucleus (MN) frequency test in bone marrow of mice: 60 mice were randomly divided into blank control group, As2O3 group and 1.2 μg/mL,2.4 μ g/mL,4.8 μ g/mL curcumine+As2O3 group (n=12).Blank control group and As2O3 group were administered salad oil through intragastrical perfusion for 7 days.Then blank control group was injected intraperitoneally with 0.5 mL saline,other three groups were injected with 0.5 mL As2O3 solution (20 μ g/mL).24 h later,mice were sacrificed and MN frequency test in bone marrow of mice was made. (2)MN frequency test in peripheral blood lymphocyte of human : human blood samples were divided into 13 groups:blank control group,1 μg/mL,2 μg/mL,4 μg/mL As2O3 group,5 μg/mL,10 μg/mL,20 μg/mL cureumine+1 μg/mL(2 μg/mL,4 μg/mL)As203 group.Then MN frequency test in peripheral blood lymphocyte of human was made to study mutagenieity of As2O3.Results: (1)Compared with As2O3 group, MN frequency in 1.2 mg/mL,2.4 mg/mL,4.8 mg/mL curcumine+As2O3 group were significantly decreased (P〈0.05 ,P〈0.01 ).(2) Compared with 1 μg/mL (2 μg/mL,4 μg/mL) As2O3 group,MN frequency in 5 μg/mL,10 μg/mL,20 μg/mL eurcum- ine+1 μ g/mL (2 μ g/mL,4 μ g/mL) As2O3 group were significantly decreased (P〈0.05,P〈0.01).Conclusion :As2O3 has dosage-correlative mutagenic effect,and curcumine has a remarkable antimutagenic effect against As2O3.
