摘要
目的:通过观察灯盏花素对体外培养的人胎儿脐动脉平滑肌细胞(Human umbilical artery smooth muscle cells)增殖的影响及球囊损伤动物模型血管内膜增殖的抑制作用,以探讨灯盏花素在再狭窄及动脉粥样硬化防治中的作用。方法:MTT法观察灯盏花素对人脐动脉平滑肌细胞增殖的抑制作用;血清生化指标检测评估灯盏花素组对血脂、肝、肾功能及脂质氧化水平等的影响;组织形态学观察球囊损伤后血管增殖。结果:MTT结果显示灯盏花素组细胞活性比率显著降低(P<0.05);血清生化指标测定结果各组间无显著性差异;组织形态学结果显示低灯盏花素组和高灯盏花素组管腔面积均显著增加(P<0.01),内膜面积显著减少(P<0.01)高、低剂量组之间无显著差异(P>0.05)。结论:灯盏花素能抑制培养的人脐动脉平滑肌细胞的增殖;灯盏花素对大鼠颈总动脉球囊损伤后血管平滑肌细胞增殖有一定的抑制作用。
Objective:This study is designed to observe the inhibitory effect of breviscapine on cultured Human umbilical artery smooth muscle cells and endangium in balloon damage Wistar rat animal model.Methods:MTT estimated cellular activity to found the inhibiting effect of breviscapine on Human umbilical artery smooth muscle cells.Serum biochemical indicator detection were measured using routine biochemical methods.Histomorphology observation.Results:MTT showed that cellular activity ration is effectively decreasing compared with control group(P〈0.05).There were no significant differences in serum among different groups(P〈0.05).Compared with control group,the intima areas was significant decreased in breviscapine high dose group and low dose group(P〈0.01).The lumens areas was significant increased (P〈0.01).there are no significant differences in the areas among different group.Conclusions:Breviscapine can inhibit the proliferation of cultural human umbilical artery smooth muscle cells;Breviscapine can inhibit the proliferation of Wistar rats common carotid artery VSMCs .
