不同降压药物对白发性高血压大鼠肾小球纤维化的影响

摘要 目的观察不同降压药物对自发性高血压大鼠(SHR)。肾小球纤维化的影响。方法将32只14周龄雄性SHR随机分为SHR对照组和治疗组(包括卡托普利组、硝苯地平组、螺内酯组),选取8只同龄雄性SD大鼠作为正常对照组。各治疗组分别给予卡托普利100mg·kg^-1·d^-1、硝苯地平30mg·kg^-1·d^-1、螺内酯30mg·kg^-1·d^-1灌胃,12周后采用免疫组化法和RT-PCR法检测肾组织转化生长因子(TGF-β1)、结缔组织生长因子(CTGF)的表达,Masson染色后计算肾组织胶原容积分数(CVF),碱水解法测定肾组织,羟脯氨酸(Hypro)含量。结果①SHR对照组CVF、Hypro含量、CTGF和TGF-β1的表达高于正常对照组,差异有统计学意义(P〈0.01);②与SHR对照组相比,卡托普利组和螺内酯组CVF、Hypro含量、CTGF和TGF-β1,的表达下降,差异具有统计学意义(P〈0.05),硝苯地平组与SHR对照组比较,无差异。③CTGF与TGF-β1、CVF和Hypro含量呈高度正相关。结论CTGF与SHR肾小球纤维化关系密切,卡托普利、螺内酯可抑制其表达,延缓高血压肾小球纤维化。 Objective To study the effects of different antihypertensive drugs on glomerular fibrosis in spontaneous hypertensive rats (SHR), and explore the possible mechanism. Methods Thirtytwo 14-week old male SHR were randomly divided into SHR eontrol group and SHR treatment groups (eaptopril group, nifedipine group, antisterone group), and 8 same age male SD rats served as normal control group. Rats in three treatment groups were administered with 100 mg· kg^-1 · d^-1 captopril, 30 mg· kg^-1 · d^-1 nifedipine and 30 mg· kg^-1 · d^-1 antisterone for 12 weeks by gavage, respectively. The renal expression of CTGF and TGF-β1 was detected by immunohistochemistry and RT-PCR. CVF was calculated after MASSON staining. The renal concentration of Hypro was determined by alkaline hydrolysis method. Results (1) As compared to normal control group , CVF and the renal concentration of Hypro and the expression of TGF-β1 and CTGF in SHR control group were significantly higher (P〈0. 01); (2) The above indices in captopril and antisterone groups were decreased as compared with SHR control group (P〈0. 05). Conclusions CTGF is closely related to glomerular fibrosis in SHR, and eaptopril and antisterone may inhibit the renal expression of CTGF, slow the advance of glomerulus fibrosis in SHR.
