THE EXPRESSION AND SIGNIFICANCE OF TGF-β1、TGF-β2 and Smad7 IN NASOPHARYNGEAL CARCINOMA

摘要 目的:探讨TGF-β/Smad信号通路的重要成员TGF-β1、TGF-β2和Smad7在鼻咽癌中的表达情况及临床病理意义。方法:采用免疫组化S-P法检测50例鼻咽癌和20例鼻咽黏膜慢性炎组织中TGF-β1、TGF-β2和Smad7蛋白的表达状况,分析其与各临床病理因素的关系。结果:①TGF-β1、TGF-β2和Smad7蛋白在鼻咽癌中的阳性表达率分别为42%、70%和40%,在黏膜慢性炎组织中的阳性表达率分别为0、100%和0,差异均有统计学意义(P<0.05);②TGF-β1与Smad7的表达与鼻咽癌的临床分期和生物学行为有一定关系(P<0.05);未发现TGF-β2的表达与鼻咽癌的主要临床病理参数相关(P>0.05);③TGF-β1与Smad7的表达存在正相关关系(rs=0.6286,P<0.05)。结论:TGF-β1、TGF-β2和Smad7在鼻咽癌的发生、发展中起一定的作用,它们有望成为辅助参考指标应用于临床,并可能为鼻咽癌的生物治疗和预后判断提供部分理论依据。 Objective: To investigate the expression and clinicopathologic significance of TGF-β1.TGF-β2 and SmadT, the important members of TGF-β/Smad signal transduction pathway in nasopharyngeal carcino ma(NPC). Methods: Expression of TGF-β1,TGF-β2 and Smad7 in tissues was detected by immunohistochemical SP method. Result: 50 NPC and 20 chronic nasopharyngitis (1)The positive expression rate of TGF-β1,TGF -β2 and Smad7 40 % respectively, nasopharyngitis tissues in 50 NPC 0, 100% and ( P d0.05). is 42%, 70% and 0 in 20 chronic (2) Expression TGF-β1 and Smad7 are correlated with tumor's clinical stage and biological behavior( P 〈0.05),but TGF-β2 is not( P 〉0.05). (3)These are positive correlation between the expression of TGF-β1 and Smad7 in NPC (rs= 0. 628 6, P 〈0.05). Conclusion: According to our study, we think TGF-β1,TGF-β2 and Smad7 take part in the carcinogenesis, progression and metastasis of NPC. They could be regarded as clinical assistant reference markers and offer some evidence for biotherapy and prognosis of NPC.
