摘要
目的从分子水平上揭示生态环境对泽泻"道地性"的影响。方法将建泽泻、川泽泻种子分别交叉在福建建瓯、四川彭山种植1年,对以上4个泽泻植株样品的18S-26S r DNA—ITS片段进行PCR扩增、测序,并运用clustalx1.83对该区序列进行分析。结果在四川栽培的建泽泻与在福建栽培的建泽泻的ITS片段序列在第614位相差1个碱基:而在福建栽培的川泽泻与在四川栽培的川泽泻序列完全一致。结论改变泽泻生态环境1年,其ITS序列发生变异的可能性很小。
Objective To reveal the effect of ecological environment on trueborn quality of rhizoma of Alisma orientale from molecular level. Methods Cloning and sequencing were applied to analyze 18S- 26S rRNA and ITS with clustalx1.83 on four species of Alisma orientale which collected from Jianou of Fujian and Pengshan of Sichuan province. Results Alignment of sequences using clustalx showed that the sequence of Jianze cultivated in Pengshan lacking of a base pairs at 614 was different from the sequence of Jianze cultivated in Jianou of Fujian province while the sequences of Chuanze cultivated in Fujian and Sichuan provinces were the same. Conclusion There was little probability of variation on the sequence of A lisma orientale which had been changed environment for one year.
