表皮生长因子对小鼠肾小管发育中细胞增殖的影响

摘要 目的:观察小鼠肾小管发育中细胞增殖和表皮生长因子(EGF)的表达特征,探讨EGF对小鼠肾小管发育中细胞增殖的影响。方法:应用5-溴-2-脱氧尿嘧啶(BrdU)标记增殖细胞,应用免疫组织化学、免疫荧光、免疫印迹技术并结合体视学方法检测生后1、3、5、6、7、14、21、28、40d小鼠肾小管BrdU阳性细胞和EGF的表达。结果:EGF于生后6d表达在肾小管上皮细胞,之后随着日龄的增加表达渐增;BrdU阳性细胞于生后1d表达,14d达高峰,之后随着日龄增加而逐渐减少;生后14d之后EGF与细胞增殖表达趋势完全相反。结论:推测EGF在生后小鼠肾发育中对肾小管的分化和成熟起重要作用,但并不促进细胞增殖。 Objective: To observe cell proliferation and the expression of EGF in renal tubules, and investigate the effect of EGF on the cell proliferation during renal tubule development in mice. Methods: BrdU was injected to label proliferative cells. Then BrdU positive ceils and EGF positive ceils were examined and analyzed by immunohistochemistry, immunofluorescence, Western blotting and stereology at neonate 1, 3, 5, 6, 7, 14, 21, 28 and 40 d of the mouse kidneys. Results: EGF immunoreactivity was found in renal tubules at Nd 6 and the expression increased following kidney development. BrdU immunoreactivity was found at Nd 1 and peaked at Nd 14 and then decreased gradually. After Nd 14, their expressive tend- ency was fully opposite. Conclusion: EGF plays an important role in regulating differentiation and maturation of renal tu- bules during mouse kidney development, but can not promote cell proliferation.
