摘要 目的探讨联合抗氧化微量营养素(AM)对脾肿瘤坏死因子(TNF)-α表达的影响。方法应用MLDS方法制备1型糖尿病(T1DM)小鼠模型,分别联合添加4种AM(Se+VE+V+Cr)和7种AM(Se+VE+V+Cr+VC+硫辛酸+烟酰胺)进行干预,利用流式细胞术和原位杂交方法观察小鼠外周血、脾淋巴细胞TNF-α表达的影响。结果联合补充AM使T1DM小鼠血糖明显降低;T1DM小鼠外周血、脾淋巴细胞TNF-α表达明显高于正常对照组,AM4组和AM7组外周血、脾淋巴细胞TNF-α表达低于T1DM模型组小鼠;T1DM模型组TNF-αmRNA表达阳性信号比正常对照组强,而AM4组和AM7组阳性信号比T1DM模型组弱。结论联合AM可下调T1DM模型小鼠脾、外周血淋巴细胞TNF-α表达,具有良好的拮抗糖尿病作用。 Objective To investigate the effect of combined antioxidant micronutrients (AM) on the TNF-α expression in spleen of T1DM mice. Methods T1DM mice models were made by multiply low dosage of streptozotocin. The mice were supplemented with four 4 sorts of AM ( Se + VE + V + Cr) or 7 sorts of AM ( Se + VE + V + Cr + VC + lipoic acid + niacinamide). The TNF-α expression in peripheral blood and spleen were observed by flow cytometry and in-situ hybridization separately. Results The supplementation of combined AM significantly decreased the blood glucose level of T1DM mice. The TNF-α expression in peripheral blood and spleen of TIDM mice was increased compared with that of control group, while the supplementation of AM decreased the expression. The TNF-α expression on protein level was consistent with that on mRNA level. Conclusions United AM could down-regulate the TNF-α expression in peripheral blood and spleen of T1DM mice, which exhibits the function against the diabetes.
