一种适宜于RT-PCR检测的辣椒病毒RNA微量提取方法 被引量:1

摘要 以感染病毒病的辣椒叶片为材料,分别用改进的Trizol试剂提取法、异硫氰酸胍提取法、改进的酚-SDS提取法提取总RNA,经过RT-PCR,利用CMV和TMV的病毒外壳蛋白设计特异引物检测。结果表明:改进的Trizol试剂提取法能获得较高质量和产量的总RNA,可以单人大批次提取,并在辣椒病毒病检测中能稳定而准确地进行规模化RT-PCR检测。 Total RNA of pepper leaves infected with virus was extracted by means of improved Trizol reagent , Guanidine-thiocyanate Reagent and Phenol-SDS respectively. The coat protein (CP) genes were amplified and tested by RT-PCR using the genomic RNA as template and the pair of specific primers from CMV and TMV. It was shown that the improved Trizol reagent method could axtract more and purer RNA, and be accurate in the pepper virus detection with RT-PCR, compared with the other methods.
