通脉口服液对实验性糖尿病肾病模型大鼠肾脏病理及转化生长因子β_1的影响 被引量:7

Effect of Tongmai Oral Liquid on Renal Pathological Feature and Tumor Growth Factor β_1 in Rats with Experimental Diabetic Nephropathy

摘要 【目的】观察通脉口服液对实验性糖尿病肾病(DN)模型大鼠肾脏病理及转化生长因子β1(TGFβ-1)的影响。【方法】选用SD雄性大鼠40例,随机分为正常对照组(8只)和造模组(32只)。正常对照组给予腹腔注射55 mg/kg无菌柠檬酸—柠檬酸钠缓冲液,饲普通饲料;造模组给予链脲佐菌素(STZ,剂量为55 mg/kg)单次腹腔注射,隔日饲高脂饲料,4周后筛选造模成功DN大鼠共21只,随机分为模型组(11只)和通脉组(10只)。通脉组给予通脉口服液(剂量为2.5.gkg-1.d-1),正常对照组及模型组给予生理盐水灌胃,饲料同前。6周后处死大鼠取肾组织,光学显微镜及电子显微镜下观察肾脏组织形态及超微结构变化,测量肾小球截面积(AMG)、细胞外基质截面积(AMM)、肾小球基底膜(GBM)厚度,计算AMM/AMG比值;同时采用免疫组化法测定肾组织TGFβ-1蛋白表达。【结果】肾组织病理形态学:通脉组系膜区增宽、基底膜增厚及肾小管上皮细胞空泡变性等均较模型组明显减轻;通脉组AMM/AMG值较模型组降低;GBM厚度较模型组变薄(均P<0.001);TGFβ-1主要定位在肾小管及间质处,肾小球表达较弱,通脉组免疫组化染色面密度及积分光密度均较模型组降低(P<0.001)。【结论】益气活血中药复方通脉口服液可能通过抑制肾组织TGFβ-1表达,减轻肾脏病理损害,发挥对DN的肾脏保护作用。 Objective To observe the effect of Tongmai Oral Liquid (TOL) on renal pathological feature and tumor growth factor β1 ( TGF-β1 ) in rats with experimental diabetic nephropathy (DN). Methods Forty SD male rats were used for the experiment. Of which, 8 were in the normal group, and the other rats were given onedose intraperitoneal injection of streptozocin ( STZ), and were fed with high-fat forage every other day. The normal group received one-dose intraperitoneal injection of aseptic citric acid-sodium citrate buffer and routine forage. After feeding for 4 weeks, the rat models of DN were randomized into the model group and TOL group, which were given normal saline and TOL (2. 5 g· kg^-1·-d^-1) respectively, and the two groups were still fed with high-fat forage. After treatment for 6 weeks, the rats were executed and the pathological features and ultrastructure of renal tissue were observed under light microscope and electron microscope respectively. Mean glomerular area (MGA), mean extracellutar matrix area ( MMA), and the thickness of glomerular basement membrane (GBM) as well as the ratio of MMA/MGA were measured. Meanwhile, the expression of tumor growth factor β1 ( TGF-β1 ) was detected by immunohistochemical method. Results In TOL group, the pathological features such as broadened mesentery, thick basement membrane and vacuolar degeneration in the renal tubular epithelium were relieved, MMA/MGA ratio was decreased, GBM became thin, and the expression of TGF-β1 was strong mainly in renal tubular and interstitial region while weak in glomeruli, and its area density and integral optimal density were decreased as compared with the model group (P 〈 0. 001 ). Conclusion TOL, which has the actions of strengthening Qi and activating blood, exerts protective effect on kidney of DN patients, and its possible mechanism is related with the inhibition of renal TGF-β1 expression and with the relief of renal pathological damage.
