钒化钠对人食管癌细胞系EC109的增殖抑制作用及机制探讨 被引量:3

摘要 目的研究钒化钠对食管癌细胞系EC109的增殖抑制作用,探讨钒化钠对其增殖抑制及cyclin D1表达的影响。方法应用MTT法测定钒化钠对食管癌细胞系EC109细胞株的生长抑制作用。用Western blot的方法检测钒化钠对细胞周期蛋白cyclin D1表达的影响。结果一定浓度的钒化钠对食管癌细胞系EC109有生长抑制作用。实验结果表明,与细胞增殖周期密切相关的cyclin D1蛋白表达降低。结论一定浓度的钒化钠能明显抑制食管癌细胞系EC109细胞株的生长,钒化钠的抗肿瘤机制可能与cyclin D1蛋白表达降低有关。 Aim To study cell growth inhibition of sodium vanadate on the esophageal cancer cell line EC109,and to explore the effects of sodium vanadate on its mechanism and expression of cyclin D1. Methods Cell growth inhibition in vitro was evaluated with MTT assay in esophageal cancer cell line EC109. Western blot analysis was applied to determine modification of cyclinD1 proteins. Results The inhibition effects of sodium vanadate on EC109 cells were signifi candy dose-dependent and time-dependent, and the expression of cyclinD1 was evidently decreased by sodium vanadate of certain Concentration. Conclusions Sodium vanadate inhibits the proliferation of human esophageal cell line EC109 significantly in a dose-dependent and time-dependent manner. The antitumor mechanism of sodium vanadate may be related to down-regulation of cyclinD1 expression.
