淋球菌IR区基因突变及mtrF表达水平与多重耐药的相关性研究 被引量:2

Correlation Between mtrF Gene Expression and Mutations of IR Region of Neisseria Gonorrhoeae and High-level Multiple Resistance

摘要 目的探讨淋球菌多传递耐药系统回文序列(IR)区基因突变及mtrF表达水平与多重耐药的相关性。方法采用纸片扩散法测定菌株的耐药性,琼脂稀释法检测菌株的耐药水平,PCR法扩增mtrR编码基因和IR区基因并进行测序分析,同时用RT-PCR测定mtrF基因的表达。结果5株敏感菌及6株仅耐红霉素的菌株mtrR和IR区基因均未发生突变,21株多重耐药菌均发生了突变(8株低~中等水平耐药株发生mtrR编码基因突变,13株高度多重耐药株IR区发生基因突变或同时存在mtrR单位点突变)。高度多重耐药株mtrF基因表达量(1.019±0.161)显著高于敏感组(0.595±0.064)(P<0.01)和低~中等水平多重耐药组(0.671±0.073)(P<0.01),而后两者间mtrF基因表达量无明显差异。结论在介导产生多重耐药过程中,淋球菌IR区基因突变及mtrF基因的高表达可能发挥着十分重要的作用。 Objective To study the relationship between mtrF gene expression and mutations of IR region of Neisseria gonorrhoeae and high-level multiple resistance. Methods The susceptibilities of isolated srtains were tested by disc diffusion method and agar dilution method. The mtrR, IR region gene and mtrR-coding sequence were amplified by PCR and directly sequenced. The mtrF expression was detected by reverse transcriptional polymerase chain reaction(RT-PCR). Results There was no gene mutation detected in mtrR-coding sequence and in IR region of 5 sensitive strains and 6 erythromyein-resistant strains. However, there were gene mutations detected in mtrR-coding sequence or in IR region of 18 multi -resistant strains. Among 21 multi-resistant strains,there were 8 strains detected mutations in mtrR-coding sequence and 13 strains detected mutations in IR region. Comparing with mtrF expression in susceptive group and inter-mediated resistant group, mtrF expression was up-regulated in high-level multiple resistant group(P 〈 0.01 ). The mtrF expression had no significant change between susceptive group and inter-mediated resistant group. Conclusion The mtrF higher expression and gene mutations in IR region may play an important role in mediating high-level multiple resistance in Neisseria gonorrhoeae.
