摘要
目的:通过研究大鼠半乳糖性白内障晶状体中氯通道蛋白-3(chloride channel protein.3,CLC-3)和水通道蛋白-1(aquaporin-1,AQP-1)表达水平的变化,探讨CLC-3和AQP-1与白内障发病的关系。方法:将半乳糖注射Wistar大鼠单侧眼球后,裂隙灯下观察晶状体混浊情况,分别在造模后不同时间点摘取模型组和对照组大鼠的眼球,取出晶状体,运用实时荧光定量PCR方法检测各组晶状体中CLC-3 mRNA和AQP-1 mRNA含量的变化。结果:透明晶状体和半乳糖性自内障晶状体中均有CLC-3 mRNA和AQP-1 mRNA的表达;生理盐水对照组各时间点CLC-3 mRNA和AQP-1 mRNA表达无统计学差异;模型组于3、7、14d时CLC-3 mRNA表达逐渐增高,均高于同期对照组(P〈0.01),到14d时约达正常水平7倍,21、30d时又有所下降,均低于同期对照组(P〈0.01);各时间点晶状体CLC-3 mRNA含量两两比较,差异均有极显著意义(P〈0.01);模型组于3d时AQP-1 mRNA表达已下降,与同期对照组比较差异有显著性意义(P〈0.05),且随着白内障进一步发展,AQP-1 mRNA含量逐渐降低,均低于同期对照组(P〈0.01);各时间点晶状体AQP-1 mRNA含量两两比较,差异均有极显著意义(P〈0.01)。结论:晶状体中CLC-3和AQP-1表达的变化可能与白内障的发生发展有着密切的联系。
Objective: To investigate the expression of chloride channel protein - 3 ( CLC-3 ) and aquaporin-1 ( AQP-1 ) gene in galactose - induced cataract and normal lens of rats, to determine whether CLC-3 and AQP-1 expression was related to the occus and progression of cataract. Methods: Galactose was injected retrobulbarly into Wistar rats. The opacity of lens was examined by slit lamp. The lens were extracted periodically from each experimental group and control group and real - time quantitative PCR techniques were used to determine the expression of CLC-3 mRNA and AQP-1 mRNA in lens during different cataract phases. Results: CLC-3 mRNA and AQP-1 mRNA were expressed in both lens of normal rats and cataract rats. During our experiment, the expression of CLC-3 mRNA and AQP-1 mRNA in lens of control group was not changeable obviously. CLC-3 mRNA in lens gragually increased at 3,7,14 days after injection of D - galactose and reached 7 - fold that of normal level at 14 days(P 〈 0.01 ). CLC-3 mRNA level decreased gradually 14 days later and was less than the normal level after 21 days (P 〈 0.01 ) . AQP-1 mRNA in lens have began to increase at 3 days after injection of D - galactose( P 〈 0.05 ) and AQP-1 mRNA level decreased persistantiy accompany to progression of cataract( P 〈 0.01 ). Conclusions: There is an overexpression of CLC-3 mRNA in early stages of galaetose induced cataract, and a downexpression of AQP-1 mRNA in the lens of eataraetous rats, which may play an importment role in the gataractgenesis.
