摘要
目的通过观察四氯化碳(CCl4)致大鼠肝纤维化过程中的白细胞介素4(IL-4)在肝组织中的表达情况,探讨肝纤维化机制。方法建立(CCl4)诱导的大鼠肝纤维化动物模型。19只Wistar大鼠分为对照组4只和肝纤维化组15只(肝纤维化组按照CCl4的作用时间又分为2、4、6、8周组)。对各组大鼠肝组织进行苏木精-伊红染色和SP法免疫组织化学染色,观察肝组织病理变化及IL-4的表达情况。结果免疫组织化学染色显示,IL-4在正常肝脏组织几乎没有表达,肝纤维化组注射CCl4诱导后,大鼠肝组织中IL-4的表达强度较对照组明显增强,主要分布在肝组织纤维化增生的汇管区。IL-4表达随CCl4作用时间延长而增强,4、6周后变化尤其明显。结论IL-4可能在肝纤维化过程中发挥着重要作用。
Objective To investigate the expression of Interleukin-4 in rat liver of hepatic fibrosis models, and further elucidate the mechanism of hepatic fibrosis. Methods The hepatic fibrosis models were established by administration of carbon tetrachloride (CC14) simultaneously for 2,4, 6,8 weeks according to A/T respectively. The liver tissues of rats in two groups(including the control group) were stained by hematoxylin-eosin and were focused on their pathological changes. The expressions of Interleukin-4 in rat liver tissues were observed with immunohistochemical staining. Results Showed by immunohistochemical staining , Interleukin-4 positive cells mainly expressed in hepatic portal area of fiber proliferation. The expressions of Interleukin-4 in the normal liver tissues was not significant. Injected by carbon tetrachloride (CC14), the expressions of Interleukin-4 were significantly increased in contrast to normal control group, and the amount of expression was gradually increased in 2,4, 6,8 weeks liver tissues, especially after 6 weeks. Conclusion Interleukin-4 might played a vital role on hepatic fibrosis.
