摘要
改进了苏云金芽胞杆菌(Bacillus thuringiensis,Bt)内生质粒提取技术,提取了Bt库斯塔克亚种、以色利亚种和一株对鳞翅目害虫有活性的野生菌株的质粒,利用琼脂糖凝胶电泳分析了质粒图谱,进一步检测了质粒的浓度与纯度。实验结果表明,该方法与传统技术相比,操作简单、耗时短,提取的质粒纯度高、条带清晰,染色体DNA污染较少。为Bt杀虫基因克隆、质粒特性分析等研究创造了有利条件。
In this report,a novel method was improved for extraction of endogenous plasmids from Bacillus thuringiensis(Bt)strains,and plasmids of 5 strains were prepared by this method,including 3 B.thuringiensis subsp.kurstaki strain(HD1,HD73 and a crystal negative strain of HD73-),a sort of B.thuringiensis subsp.israelensis strain(Bti)and B.thuringiensis isolate S6 with lepidopteran insecticidal activity.The concentration,the purity and the patterns of the plasmids were analysed.The new method can not only save time,but also produce purer plasmids with clearer patterns,and decrease contamination from chromosomal DNA.This study creates the advantage for the research on cloning of pesticidal genes and characterization of endogenous plasmid from B.thuringiensis.
