摘要
目的:用高效液相色谱-质谱(HPLC-MS)法研究使药降香对君药丹参中丹参素兔药代动力学及组织分布的影响.方法:加热回流法制备丹参单煎液及丹参-降香合煎液,以4-羟基苯甲酸为内标,丹参素为指标,HPLC-MS法测定不同采样点丹参素的血药浓度及组织中的含量,DAS2.0数据处理软件拟合房室模型并计算药代动力学参数.结果:配伍前,丹参素的t1/2α为20.8min;V1为84.5L/kg;AUC(0~∞)为119.4mg/(L.min);CL为0.42L/(min.kg).配伍后,丹参素的t1/2α为30.9min;V1为108.3L/kg;AUC(0~∞)为178.7mg/(L.min);CL为0.28L/(min.kg).降香能使丹参素组织分布特征由肾>心>脑>肝变为肾>肝>脑>心.结论:配伍后,使药降香能提高君药丹参中丹参素吸收入血的程度,使丹参素向肝、脑中转移,为阐明"君-使"对药"丹参-降香"的相使作用提供依据.
AIM: To investigate the effect of Shi herb (Dalbergia odorifera, DO) on the pharmacokinetic parameters and tissue distributed profile of Danshensu (DSS) in Jun herb ( Salvia miltiorrhiza, SM). METHODS: The total extracts of SM and SM coupled with DO were prepared by reflux method. With 4- hydroxybenzoic acid as an internal standard, the concentration of DSS in plasma and tissue samples were determined by high performance liquid chromatography-trap mass (HPLC-MS) spectrometry. Finally, the pharmacokinetic parameters were calculated by the software of DAS 2.0. RESULTS : After oral administration of SM, the pharmacokinetic parameters of t1/2=, V1, AUC(0→∞)〉 and CL were 20.8 min, 84.5 L/kg, 119.4 mg/( L · min) and 0.42 L/(min · kg), respectively. After oral administration of SM coupled with DO, the pharmacokinetic parameters of t1/2,, V1 , AUC(0→∞)〉 and CL were 30.9 min, 108.3 L/kg, 178.7 mg/(L · min) and 0.28 L/( min · kg) , respectively. Moreover, the characteristic profile of DSS tissue distribution were changed from kidney 〉 heart 〉 brain 〉 liver to kidney 〉 liver 〉 brain 〉 heart. CONCLUSION: After oral administration of SM coupled with DO, the content of DSS in SM in the plasma could be increased by DO, and the concentration of DSS in the tissue of liver and brain could also be increased by DO, providing the investigation for elucidating the synergistic interaction of Jun-Shi coupled herbs (i.e. SMandDO).
