摘要
以中嘉8号杨试管苗叶片为外植体,研究了预培养时间、菌液浓度、共培养时间和选择方式等因子在农杆菌介导的杨树基因转化过程中对提高转化率的影响。结果表明:共培养4d、延迟10d选择和延迟20d选择的3个处理Kmr芽的平均诱导频率分别为20%,28.9%和26.7%,显著优于其它因子。因此,影响杨树转化率的主要因素是选择方式,其次是共培养时间。
In the experiment, the leaves from Populus dehoides ( I - 69× I - 63 ) in vitro were used as explants and several factors such as Agrobaeterium consistence, pre - culture time, co - culture time and selecting method were studied for improving transformation rate of BtCryl A in the poplar. Among the factors, the transformation rate can reach 20% when 4 days of co - culture, 28.9% and 26.7% when selection were delayed after 10 days and 20 days of co - culture. Thus, the first factor is selecting method, the second is co - culture time.
