大肠腺癌组织中p57^(kip2)蛋白和基因的表达及其与临床病理的关系

Expression of p57^(kip2) Protein and p57^(kip2) mRNA in Human Colorectal Adenocarcinoma and Their Clinicopathological Correlation

摘要 目的探讨细胞周期调控抑制因子p57kip2蛋白和基因在大肠腺癌发生发展中的作用。方法采用免疫组化技术SP法,检测p57kip2蛋白在37例大肠腺癌和26例正常大肠黏膜组织中的表达;采用RT-PCR技术,检测p57kip2基因mRNA在32例大肠腺癌和癌旁组织中的表达。结果p57kip2蛋白阳性表达率在大肠腺癌组织中为40.5%,显著低于正常大肠黏膜组织(χ2=5.039,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=8.914,χ2=4.416,P<0.05);p57kip2基因mRNA阳性表达率在大肠腺癌组织中为34.4%,显著低于癌旁大肠黏膜组织(χ2=4.016,P<0.05),并与大肠腺癌组织分化程度、淋巴结转移无关;大肠腺癌组织p57kip2基因mRNA表达水平显著低于癌旁组织(P<0.05)。结论p57kip2蛋白的低表达与大肠腺癌的发生发展有关,p57kip2基因mRNA的低表达与大肠腺癌的发生有关,且p57kip2基因mRNA的低表达可能是p57kip2蛋白低表达的原因之一。 Objective To investigate the effects of inhibiting factor of cell cycle regulation- p57^kip2 protein and gene in the genesis and progression of human colorectal adenocarcinoma. Methods The expressions of p57^kip2 protein in tumor tissues of 37 patients with colorectal adenocarcinoma and in normal colorectal mueosal tissues of 26 subjects were detected by SP immunohistochemical technique. The expressions of p57^kip2 gene mRNA in tumor tissues and adjacent colorectal tissues of 32 patients were detected by RT- PCR. Results p57^kip2 protein positive-expression rate in tumor tissues of colorectal adenocareinoma was 40. 5%, which was lower than that in normal colorectal mucosal tissues (χ^2 = 5. 039, P〈0.05), p57^kip2 protein positive-expression correlated significantly with tumor cell differentiation and lymph node metastasis (χ^2 = 8. 914,χ^2 = 4. 416,P〈0. 05) ; p57^kip2 gene mRNA positive-expression rate in tumors of colorectal adenocarcinoma was 34. 4%, which was lower than that in adjacent colorectal mucosal tissues (χ^2 = 4. 016,P〈0. 05), and was not correlate remarkably with tumor cell differentiation, lymphy node metastasis; the expression level of p57^kip2 gene mRNA in tumor tissue was lower than in adiacent corlorectal mucosal tissues (P〈0. 05). Conclusion The decreased expression of p57^kip2 protein may play an important role in the genesis and progression of colorectal adenocarcinoma. The decreased expression of p57^kip2 gene mRNA may contribute to the occurrence of colorectal adenoearcinoma. The decreased expression of p57^kip2 protein may relate with the decreased expression of the expressions of p57^kip2 gene mRNA.
