摘要 本文建立了反相高效液相色谱法测定Levofloxacin(左旋氧氟沙星)的血药浓度方法,样品经液-液提取后,C(18)柱上分析,流动相为甲醇:0.01M磷酸盐缓冲液:0.5M四丁基溴化铵,紫外检测波长294nm,线性范围0.5~4.0μg/ml,平均回收率为97.17%,日内误差<7%,日间误差<8%,本法具有简便、灵敏、快速、准确的优点,已用于药代动力学的研究。 A RP-HPLC method for determination of levofloxacin in serum was developed.The sample was extracted by one step liquid-liquid extraction followed by separation on C18 column with a mobile phase of mixture of methanol:0.01M phosphate buffer:0.5M tetrabutyl ammonium bromide.The UV detector wavelength was set at 294nm.The standard curve was linear in 0.5~4.0μg/ml range.The average recovery was 97.17 % and the variations of inter-day and intra-day were less than 7% and 8%, respectively.The method is simple,sensitive,accurate and has been applied to pharmacokinetic studies in human.
