摘要
本研究利用[3H]-TdR掺入法观察不同浓度的氟化钠及中药续断组分对体外培养的正常成人成骨细胞的影响。结果显示,氟化钠能直接影响成骨细胞的增殖,且具有双向调节效应,在一定浓度范围内(1.25~10μmol/L)能促进细胞增殖,但其有效浓度范围相对狭窄,浓度过高(>20μmol/L)反而抑制成骨细胞的生长。中药续断的两种组分对成骨细胞的增殖均具有促进作用,但有各自不同的有效浓度范围。本研究提示氟化钠对成骨细胞具有直接的作用。中药续断对成骨细胞也有促增殖作用,理论上有望从促骨形成方面用于骨质疏松的预防及治疗,同时也为中药的筛选提供了一条新路。
n order to further understand the effect of fluoride on the growth of osteoblasts cultured in vitro, and to find out a more effective agent for the treatment of osteoporosis, the authors studies the effect of different concentrations of sodium fluoride and dipsacus asperoides on normal osteoblast in vitro proliferation with the method of incorporation of tritiated thymidine into DNA.The results showed that, sodium fluoride had a double effect on osteoblast proliferation. With the dose of 1.25 to 10 μmol/L, there was significant enhancement of thymidine incorporation into human osteoblast. In contrast, at 20 μmol/L or higher, an inhibitory effect was observed. The 95%EtOH extracts of dipsacus asperoides had a dose-dependent stimulating effect on the DNA synthesis in the osteoblast, however the effect was observed only at high concentration of 100 μg/ml. The result suggests that, sodium fluoride has a direct stimulative effect on human osteoblast proliferation at certain concentrations. The maximal effect appeared at a concentration close to the toxic levels, the extracts of dipsacus asperoides were also effective for stimulating osteoblast proliferation and it may possible be a useful agent for the treatment of osteoporosis.
