摘要 采用静态离子交换法从鸡蛋清中提取溶菌酶,探讨不同条件对溶菌酶收率等的影响,确定了最优操作参数。所用树脂为D152阳离子交换树脂。最佳提取条件为:树脂用量占蛋清体积的30%;吸附时间6h;去离子水洗涤杂蛋白3次;用10%硫酸铵溶液洗脱3次,洗脱时间1h;用洗脱液体积2倍的丙酮沉淀溶菌酶。试验从300mL蛋清中制得溶菌酶0.896g,得率0.295%,比酶活可达9500U/mg左右,纯度为96.3%。 In this paper,lysozyme was extracted from egg white by using static ion-exchange method. The process conditions for abstraction lysozyme were discussed in detail. And the optimal operating parameters were established. The resin used in this experiment was cation-exchange chromatography (D152). The optimal condition was as follows:30% volume ratio of resin in egg white;6 hours absorption time;washes 3 times by de-ionized water to remove the impurity protein;elute 3 times by 10% ammonium sulfate solution for 1 hour;deposit lysozyme by acetone which is 2 times volume than that of elution. 300 mL egg white could produce 0.896 g lysozyme with 9 500 U/mg activity,0.295% yield and 96.3% purity.
