摘要
为探讨免疫组织化学染色中常用的PicTureTM二步法与链霉素抗生物素-过氧化物酶(streptaridin-per-oxidase,SP)法在中枢神经下丘脑中催产素定位的优劣,选取摩拉杂交一代水牛(试验组)与广西本地水牛(沼泽型)(对照组)的下丘脑分别进行染色,结果显示:PicTureTM二步法试验组的平均直径(P:0.015)和阳性面积(P:0.02)均差异显著,光密度值未见显著差异。SP法光密度、平均直径和阳性面积均未见显著差异。SP法光密度值(0.913 9±0.034 87)明显高于二步法光密度值(0.213 9±0.056 42),可能与DAB染色时间有关。就本试验而言,从背景非特异性染色、阳性面积和平均直径差异性等方面考虑,在神经内分泌神经元定位中,二步法优于SP法。
In the present study, the immunohistochemical streptaridin - peroxidase (SP) method and PicTure^TM two - step method were used to study the effects of the two immunohistochemical methods on determining the expression of oxytocin in hypothalamus of swamp type water buffalo and murrah hybrid water buffalo. The results showed that the mean diameter(P:0.015 ) and masculine area(P:0.02) of murrah hybrid water buffalo changed significantly in the two - step method treatment, but the mean density did not change. The mean density(0.913 9 +0.034 87) in the SP method treatment was significantly larger than that in the two - step method(0.213 9 +0.056 42) ,which was probably affected by staining. According to the non -specificity staining and the variability of mean diameter and masculine area, PicTureTM two - step method gains an advantage in the study of neuroendocrine.
