摘要
【目的】研究大鼠脊髓全横断后脑源性神经营养因子(BDNF)在小脑前叶皮质表达的变化,为进一步探索脊髓损伤后的再生修复机制和神经营养因子治疗脊髓损伤提供实验依据。【方法】雌性SD大鼠72只,随机分为3组:正常对照组,假手术组和脊髓全横断组,分别以脊髓全横断组术后1,3,7及21 d为标准时间点每组随机取6只大鼠收集小脑前叶皮质标本,采用免疫组织化学ABC染色法和图像分析技术检测BD-NF在不同组别小脑前叶皮质表达变化情况。【结果】脊髓全横断组中,BDNF在小脑前叶皮质的表达在1 d组,3 d组尚未出现明显增加,到7 d组时表达则明显升高,21 d组时表达仍高于正常对照组和假手术组。【结论】脊髓全横断后,BDNF在小脑前叶皮质表达的上调可能与神经元的保护及修复有关。
[Objective] To investigate the changes of brain-derived neurotrophie factor(BDNF) expression in the cortex of rat anterior lobe of cerebellum following the complete spinal cord transection in order to provide the experimental evidence for the mechanisms of regeneration and repair after spinal cord injury and for neuro- trophins to treat spinal cord injury. [Methods]Female Sprague-Dawley rats were divided into three groups at random ; (1) Normal control group ; (2)Sham operation group ; ( laminectomy only) ; (3)Transected spinal cord inj u ry group. The samples were collected at the four different time points. Then the expression of BDNF: in the cortex of anterior lobe of cerebellum was examined by immunohistoehemistry and image analysis. [Results] In the transected spinal cord injury group, BDNF expression in the cortex of anterior lobe of cerebellum in 1 day and 3 days has not yet been apparently higher, but in 7 days, the expression has been significantly higher, and in 21 days, the expression has been still higher than that in the normal control group and the sham operation group. [Conclusion]After complete spinal cord transection, the expression of BDNF in the cortex of anterior lobe of cerebellum is increased. It is possible that BDNF plays an important part in the protection and repair of the neuron.
