四氯化碳致肝硬化大鼠高同型半胱氨酸水平和PCNA基因表达升高的实验研究

摘要 目的研究肝硬化对同型半胱氨酸(HCY)水平以及主动脉血管增殖细胞核抗原(PCNA)基因表达的影响.方法肝硬化的大鼠通过CCl4诱导产生.血浆HCY检测用酶免疫分析法;胱硫醚β合成酶和PCNA基因表达测定采用RT-PCR.结果与对照组相比,肝硬化的大鼠血浆HCY水平显著升高,并与血浆III型胶原蛋白、透明质酸水平呈相关性.血浆肝组织胱硫醚β合成酶基因表达和酶活性均显著下降.在肝硬化的大鼠中,主动脉血管PCNA基因表达显著升高.结论肝硬化导致血浆HCY升高是由于HCY代谢受损所致,肝硬化可能增加动脉粥样硬化的发病机率. Objective To study the effects of liver cirrhosis on the level of homocysteine (HCY) in plasma and gene expression of proliferating cell nuclear antigen (PCNA) in aortia. Methods CC14 was used to induce liver cirrhosis in rats. The level of plasma HCY was measured using enzyme-immunoassay. The mRNA levels of cystathione-synthase and PCNA were detected by RT-PCR. Results The level of plasma HCY in rats with hepatic cirrhosis was significantly higher than that of control. A correlation analysis revealed that the HCY level was significantly correlated to the levels of type III procollagen and hyaluronic acid. Both the mRNA level and enzyme activity of cystathione-synthase in rats with hepatic cirrhosis markedly decreased. The mRNA level of PCNA significantly increased, compared with that of control. Conclusions Liver cirrhosis leads to an increase in the level of plasma HCY, due to insult in HCY metabolism. Liver cirrhosis may increase the rate of atherosclerosis.
