摘要
应用电子克隆的方法获得了玉米的一个Pti1(Pto-interacting 1)同源基因的全长cDNA,命名为ZmPti1-1(Gen-Bank接受号为EF158035)。推断ZmPti1-1蛋白含有362个氨基酸,分子量为39.00 kDa,pI为8.14,包含蛋白激酶催化结构域保守的11个亚结构域。在水杨酸(Salicylic acid,SA)、甘露醇、氯化钠、脱落酸(Abscisic acid,ABA)和4℃低温的诱导下,ZmPti1-1在玉米幼苗叶片中的表达量明显上调。同时,在玉米不同发育时期、不同组织器官内,ZmPti1-1的表达水平也有很大差异,在子房中表达量最高,在雄蕊中检测不到表达。试验结果表明,ZmPti1-1是一个玉米类Pti1基因,响应生物胁迫和非生物胁迫的诱导。
One full-length cDNA encoding a Pti1 (Pto-interacting 1 ) homologue was isolated from maize through in silico cloning and named as ZmPti1-1. The predicted ZmPti1-1 protein has 362 amino acids with an estimated molecular mass of 39.00 kDa and an isoelectric point of 8.14. The putative kinase catalytic domain of ZmPti1-1 contains the 11 subdomains that are typical in protein kinase. ZmPti1-1 expression was up-regulated obviously by salicylic acid, mannitol, NaCl, absicic acid and low temperature(4℃ ) in young leaves of maize. Furthermore, ZmPti1-1 displayed different expression patterns in different growth stages and tissues and was most abundant in ovaries, but was undeteetable in tassels. These results demonstrated that ZmPti1-1 is a maize Pti1-like gene and responds to biotic and abiotic stress signals.
