穴位贴敷天灸透皮吸收剂对哮喘豚鼠血清肿瘤坏死因子α的影响 被引量:15

摘要 背景:中国古代天灸疗法与现代中药经皮控释给药系统相结合研制而成的透皮吸收剂,经皮肤给药除了能起到局部作用外,还可以透过皮肤进入血液循环,发挥全身作用。目的:观察穴位贴敷天灸透皮吸收剂对哮喘豚鼠血清肿瘤坏死因子α水平的影响。设计、时间及地点:随机对照动物实验,于2007-10/2008-09在南方医科大学中药制剂实验室、广州中医大学动物实验中心完成。材料:48只豚鼠随机分为正常对照组、模型对照组、天灸透皮吸收剂药贴组及地塞米松组,每组12只。方法:用卵蛋白致敏法制作实验性豚鼠过敏性哮喘模型,诱喘成功后,天灸透皮吸收剂药贴组在哮喘发作次日开始进行穴位贴敷治疗。穴位选择:大椎穴、肺俞(双)穴、肾俞(双)穴。每次敷药6h,隔日治疗1次,共治疗7次。地塞米松组采用腹腔注射地塞米松0.5mg/kg,治疗时间与次数同天灸透皮吸收剂药贴组。模型对照组和正常对照组不给予任何干预治疗。主要观察指标:在末次诱喘后2~4h内,采用酶联免疫吸附法测定各组豚鼠血清肿瘤坏死因子α水平。结果:24只豚鼠进入结果分析,每组6只。天灸透皮吸收剂药贴组、地塞米松组豚鼠血清肿瘤坏死因子α水平明显低于模型对照组(P〈0.05):天灸透皮吸收剂药贴组、地塞米松组豚鼠血清肿瘤坏死因子a水平稍高于正常对照组(P〈0.05):天灸透皮吸收剂药贴组与地塞米松组豚鼠血清肿瘤坏死因子α平相当,差异无显著性意义(P〉0.05)。结论:穴位贴敷天灸透皮吸收剂药贴与地塞米松均能降低哮喘豚鼠血清肿瘤坏死因子α水平,从而减轻免疫炎性反应及气道高反应性。 BACKGROUND: The cutaneous permeable agent is developed by combination of ancient Chinese medicinal vesiculation therapy and transdermal controlled release system of modem Chinese medicine. Besides local action, transdermal drag delivery also has general action by entering into the blood circulation. OBJECTIVE: To observe the effect of point-application cutaneous permeable agent of crude herb moxibustion on the serum level of tumor necrosis factor alpha (TNF- α ) in guinea pigs with asthma. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at Preparation Laboratory of Traditional Chinese Medicine of Southern Medical University and Experimental Animal Center of Guangzhou University of Traditional Chinese Medicine from October 2007 to September 2008. MATERIALS: A total of forty-eight guinea pigs were divided into four groups, with 12 in each group: normal control, model control, cutaneous permeable agent of crude herb moxibustion and dexamethasone. METHODS: Guinea pigs in the model control, cutaneous permeable agent of crude herb moxibustion and dexamethasone groups were subject to establish the models of allergen induced asthma by ovalbumin sensitization. On the second day after an asthma attack, animals in the cutaneous permeable agent of crude herb moxibustion group were treated with acupoint application at the Dazhui, Feishu, and Shenshu. The animals were treated 6 hours once every other day 7 times. Animals in the dexamethasone group were administered (i.p.) 0.5 mg/kg dexamethasone. The normal and model control groups did not receive any acupoint application or injection. MAIN OUTCOME MEASURES: At the interval of 2-4 hours after the last asthma attack, the serum levels of TNF-α in the animals were measured with enzyme-linked immunosorbent assay (ELISA). RESULTS: A total of 24 guinea pigs with 6 in each group were enrolled in the final analysis. The serum levels of TNF- α in the cutaneous permeable agent of crude herb moxibustion and dexamethasone groups were significantly lower than that in the model control group, and significantly higher than that in the normal control groups (both P 〈 0. 05). There was no difference between the cutaneous permeable agent of crude herb moxibustion and dexamethasone groups (P 〉 0.05). CONCLUSION: The point-application cutaneous permeable agent of crude herb moxibustion, similar to dexamethasone injection, can decrease the serum TNF- α level, so as to reduce the immunity inflammatory reaction and airway hyperactivity.
