摘要
目的探讨建立紫杉醇致大鼠外周神经病理性疼痛模型的可行性。方法40只成年雄性SD大鼠随机分为4组,各组均在第1、3、5、7天腹腔注射紫杉醇,剂量分别为0、0.5、1.0、2.0mg/(kg·d),分别于用药前和用药后第3、5、7、12、16、20和25天测定大鼠的机械缩足反射阈值(MWT)和热缩足潜伏期(TWL),用药后第20天采用透射电镜观察坐骨神经超微结构。结果腹腔注射紫杉醇1.0mg/kg组大鼠最早在用药的第5天出现机械痛敏和热痛敏,机械痛阈在用药的第12天达到最小,而热痛阈在用药的第16天达到最小。腹腔注射紫杉醇1.0mg/kg组大鼠的坐骨神经纤维髓鞘板层结构松散、肿胀变形、排列紊乱、厚薄不均。结论使用SD大鼠重复腹腔注射1.0mg/kg紫杉醇的方法可以成功建立紫杉醇致大鼠外周神经病理性疼痛模型。
Objective To investigate the feasibility of establishing an animal model of peripheral neuropathic pain induced by paclitaxel in rats. Methods Forty adult male SD rats were randomly divided into 4 groups (n= 10 in each group). The rats were injected intraperitoneally on 4 alternate days (days 1, 3, 5 and 7) with vehicle or paclitaxel (0.5, 1.0 or 2.0 mg/kg). Mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) of rats' hind paw were measured before and 3, 5, 7, 12, 16, 20 and 25 days after injection. The ultrastructures of sciatic nerve were observed under an electronic micro scope on the 20th day of paclitaxel injection. Results In paclitaxel groups, mechanical allodynia and heat-hyperalgesia appeared on the 5th day after paclitaxel (1 mg/kg) injection, and MWT and TWL were decreased to minimum on the 12th day and 16th day after paclitaxel injection respectively. Obvious demyelinated procedure in sciatic nerve was observed in paclitaxel (1 mg/kg) groups, accompanied by edema and disordered array of myelin. Conclusion An animal model of paclitaxel-induced painful peripheral neuropathy in the rats was successively established by intraperitoneally injecting on 4 alternate days with 1 mg/kg paclitaxel.
