摘要
在成功建立大豆下胚轴悬浮培养细胞体系的基础上,研究了茉莉酸甲酯、二甲亚砜(DMSO)和苯丙氨酸对大豆悬浮细胞生长状态、总异黄酮含量及大豆素含量的影响.结果表明:DMSO和苯丙氨酸加入大豆悬浮培养体系后有利于悬浮细胞生长,对悬浮细胞干物质积累有一定的促进作用.75 mmol/L DMSO加入悬浮体系培养48 h时,悬浮细胞中总异黄酮和大豆素的积累显著增加,分别为对照的1.6和1.8倍;10μmol/L的苯丙氨酸处理悬浮细胞48 h时,不仅能够促进悬浮细胞中大豆素的积累,同时还有利于提高染料木素的含量;而加入茉莉酸甲酯则易使大豆悬浮细胞褐化,对悬浮体系中总异黄酮及大豆素的积累均没有明显作用.
The effects of methyl jasmonate,dimethylsulfoxide (DMSO) ,and phenylalanine on the growth of suspended cells,accumulation of total isoflavones and daidzein in cell-suspension cultures of soybean were studied. Cell-suspension cultures of soybean were established from soybean hypocotyl callus and could accumulated isoflavone stably. The results showed that dimethylsulfoxide and phenylalaninc could promote cell growth and dry matter accumulation. The contents of total isoflavone and daidzein in cell-suspension cultures of soybean were greatly increased when 75 mmol/L dimethylsulfoxide was added in the culture system for 48 h,being 1.6 and 1.8 times of the control, respectively. Methyl jasmonate, however,could not contribute to the growth of the suspened cells and elicit the accumulation of total isoflavone and daidzein in cell-suspension cultures of soybean.
