灰斑古毒蛾核型多角体病毒EcoRⅠ-U片段的克隆与序列分析

摘要 【目的】分析灰斑古毒蛾核型多角体病毒(Orgyia ericae nucleopolyhedrovirus,OrerNPV)EcoRⅠ-U片段分子生物学特征,为研究其分子特性及其在OrerNPV感染周期中的功能提供科学依据。【方法】克隆、分析OrerNPV EcoRⅠ-U片段所包含的若干基因,并应用生物软件对OrerNPV泛素基因(Ubi基因)进行了进化分析。【结果】OrerNPV EcoRⅠ-U片段包含Ubi基因、AcORF34同源基因和39K基因部分序列,在该区域中编码Ubi基因的开放阅读框(ORF)由246个核苷酸组成,可以编码81个氨基酸,预汁蛋白质分子质量为8.9ku。OrerNPV Ubi基因与苜蓿丫纹夜蛾核型多角体病毒(AcMNPV)、家蚕核型多角体病毒(BmNPV)、薄荷灰夜蛾核型多角体病毒(RaouM-NPV)的同源性最高,均达92%,与小菜蛾颗粒体病毒(Plutella xylostella granulovirus,PxGV)的同源性最低,为62%。OrerNPV与AcMNPV、BmNPV和RaouMNPV的亲源关系最近。EcoR Ⅰ-U片段+M13端含有编码39K基因(pp31)的部分序列,该多肽与10种NPV的39K序列有48%~52%的同源性。【结论】明确了OrerNPV EcoRⅠ-U片段的序列特征及OrerNPV Ubi在泛素蛋白家族中的进化关系。 [Objective] Molecular characteristics were analyzed on Orgyia ericae nueleopolyhedrovirus (OrerNPV). [Method] Here, the cloned OrerNPV EcoRⅠ-U fragment in which the Ubi and homologues of AcORF34 and 39K were found was analyzed. [Result] The Ubi open reading frame was 246 nucleotides long that coded a peptide of 81 amino acids with a predicted molecular weight of 8.9 ku. The peptide of OrerNPV Ubi shared a highest similarity of 92% to Autographa ealifornica multiple nucleocapsid nucleopolyhedrovirus,Bornbyx mori nucleopolyhedrovirus and Rachiplusia ou multiple nucleocapsid nucleopolyhedrovirus;while it shared a lowest similarity with Plutella:rylostella granulovirus GV of 62%. The phylogenetic results of Ubi based on 21 species showed that AcMNPV,BmNPV and RaouMNPV were mostly related to OrerNPV. The 39K amino acid residues in the M13+ terminal of OrerNPV had 48%-52% identities with homologous genes from other 10 NPVs. [Conclusion] The result clarified the characteristics of OrerNPV EcoR Ⅰ-U fragment and phylogenetic relationships of OrerNPV Ubi gene in the Ubi family.
