摘要
目的探讨变异链球菌生物膜成熟初期可溶性蛋白表达情况。方法采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)技术,分析变异链球菌生物膜成熟初期不同时间点(16、18、20、22、24h)菌体可溶性蛋白的表达情况,并通过光学显微镜观察各时间点菌斑生物膜的形态和结构特征。结果镜下见16h形成的生物膜主要由散在微菌落构成,随时间延长其菌落逐渐变大并与相邻菌落融合(18~22h),最终相互重叠(24h)。比较5个时间点变异链球菌生物膜中菌体可溶性蛋白未见特异表达的蛋白条带,且相同位置的蛋白条带在蛋白表达量上差异无统计学意义。结论本研究条件下,可观察到变异链球菌菌落从聚集到重叠形成成熟生物膜的过程,16~24h成熟初期变异链球菌生物膜中菌体可溶性蛋白表达差异无统计学意义。
Obejective To investigate Streptococcus mutans (S.mutans) biofilm protein expression at early mature stages. Methods The cell soluble protein expression of S.mutans biofilm was measured at different time points (16, 18, 20, 22, 24 h) respectively by SDS-polyacrylamide gel electrophoresis (SDS-PAGE). The morphology and structural characteristics of biofilm were also observed under optical microscope. Results 16 h after cultivation, the biofilm mainly consisted of micro-colonies. Subsequently the colonies expanded and integrated with each other (18- 22 h), and eventually develop into overlapping (24 h). No specific protein bands were observed in S.mutans biofilm soluble protein expression products at different time points. There were also no significant differences in quantity of protein expression. Conclusion In this study, S.mutans colonies were observed to be incling to integrate and overlap. There were no significant differences in S.mutans biofilm cell soluble protein expression at 16- 24 h early mature stages.
