摘要
目的研究荞麦花叶总黄酮(TFBFL)对去甲肾上腺素(NE)所致大鼠心肌肥厚的保护作用并探讨其可能机制。方法腹腔注射NE建立大鼠心肌肥厚模型,以TFBFL100,200,400mg/(kg·d)和阳性药物卡托普利(CAP)50mg/(kg·d)灌胃给药15d。计算左心室肥厚指数(LVWI:LVW/BW),和心重指数(HWI:HW/BW,LVW/HW)。采用HE染色法,观察心肌组织及其间质的组织形态学变化。并取大鼠左室心肌上清液用分光光度法检测血管紧张素Ⅰ转化酶(ACE)含量。结果与肥厚模型组比,TFBFL能降低LVWI和HWI,改善心肌病理变化。且可不同程度的降低压力负荷后左室心肌ACE的含量,400mg/(kg·d)组与阳性药物CAP组治疗效果相近。结论TFBFL对NE所致大鼠心肌肥厚具有逆转保护作用,其机制可能是拮抗了ACE。
Objective To study the protection of total flavones of buckwheat flower and leaf against cardiac hypertrophy induced by norepinephrine(NE)and its mechanism. Methods We established the cardiac hypertrophy models of rats by i.p. Meanwhile, the rats were administrated TFBFL and Capropril by i. g at the dose of 100,200,400mg/(kg ·d)and 50mg/(kg · d)for 15 days. LVMI and HWI were measured. The ACE content of myocardium was measured by colorimetry. Myocardium and it's stroma were observed through HE staining. Results Compared with hypertrophy group, TFBFL could decreased LVMI and HWI. And it also could improve cardiac pathological changes, could inhibit ACE. The curative effects of TFBFL(400mg/kg · d) was similar with captropril. Conclusion TFBFL has some protection on NE- induced cardiac hypertrophy. The mechanism may be related to inhibition of ACE.
