摘要
目的探讨CyclinD1、P21WAF1/CIP1和Survivin与脑星形细胞瘤的相关性。方法采用ElivisionTMplus免疫组化方法,检测10例正常脑组织、60例脑星形细胞瘤中CyclinD1、P21WAF1/CIP1和Survivin的表达情况。结果在正常脑组织和脑星形细胞瘤Ⅰ级、Ⅱ级、Ⅲ级、Ⅳ级中的阳性表达情况:(1)CyclinD1阳性表达率分别为:20.00%(2/10)、25.00%(4/16)、43.75%(7/16)、50.00%(7/14)、71.43%(10/14),其表达与病理分级呈显著正相关(r=0.357,P<0.01);(2)P21WAF1/CIP1的阳性表达率分别为:0(0/10)、81.25%(13/16)、62.50%(10/16)、42.86%(6/14)、27.27%(3/14),其表达与病理分级呈显著负相关(r=-0.548,P<0.001);(3)Survivin的阳性表达率分别为:0(0/10)、56.25%(9/16)、68.75%(11/16)、92.86%(13/14)、100.00%(14/14),其表达与病理分级呈显著正相关(r=0.539,P<0.005);(4)CyclinD1表达阳性的28例中,P21WAF1/CIP1阳性表达11例,阳性率39.29%(11/28),Cy-clinD1表达阴性的32例中,P21WAF1/CIP1阳性表达21例,阳性率65.63%(21/32),两者呈负相关(r=-0.362,P<0.02);(5)Survivin表达阳性的47例中CyclinD1阳性表达27例,阳性率57.45%(27/47),Survivin表达阴性的13例中CyclinD1阳性表达1例,阳性率7.69%(1/13),两者呈显著正相关(r=0.411,P<0.005);(6)P21WAF1/CIP1表达阳性的32例中survivin阳性表达20例,阳性率62.50%(20/32),P21WAF1/CIP1表达阴性的28例中Survivin阳性表达27例,阳性率96.43%(27/28),两者呈显著负相关(r=-0.411,P<0.005)。结论(1)CyclinD1过表达、P21WAF1/CIP1表达减弱或消失以及Survivin的高表达与脑星形细胞瘤的发生、发展有关;(2)联合检测三者在一定程度上反映脑星形细胞瘤的生物学行为。
Objective To discuss the dependability of CyclinD1. P21^WAF1/CIP1 and Survivin in human astrocompass. Methods ElivisionTMplus immunohistochemical staining method was used to detect the expression of CyclinD1. P21^WAF1/CIP1 Survivin in 60 human astrocytoma. Results The expression status in normal brain tissue and astrocytoma of grade Ⅰ .grade Ⅱ .grade Ⅲ and grade Ⅳ,(1) The CyclinD1 positive ratios were 20. 00% (2/10), 25. 00% (4/16), 43. 75% (7/16), 50. 00% (7/14), 71. 43% (10/14), there was significant positive correlation difference between four grades(r=0. 357,P〈0.01). (2)The P21^WAF1/CIP1 positive ratios were 0(0/10),81.25%(13/16),62.50%(10/16) ,42.86%(6/14),27.27%(3/14) ,there was significant negative correlation difference between four grades (r = - 0. 548, P〈 0. 001). (3) The Survivin positive ratios were 0(0/10),56.25%(9/16),68.75%(11/16),92.86%(13/14),100.00%(14/14), there was significant positive correlation difference between four grades(r=0. 539, P〈0. 005). (4)In 28 example positive expression of CyclinD1, there were 11 examples showing positive expression of P21^WAF1/CIP1 , the positive ratio was 39.29%(11/28) ,and in 32 example negative expression of Cy- clinD1, there were 21 examples showing positive expression of P21^WAF1/CIP1 , the positive ratio was 65. 63%(21/32), there was negative correlation difference between CyclinD1 and P21^WAF1/CIP1 (r=-0. 362,P〈0.02). (5)In 47 example positive expression of Survivin ,there were 27 examples showing positive expression of CyclinD1, the positive ratio was 57.45 % (27/47), and in 13 examples showing negative expression of Survivin, there are 1 example showint positive expression of CyclinD1, the positive ratio was 7.69% (1/13), there was significant positive correlation difference between CyclinD1 and Survivin(r= 0. 411, P〈0. 005). (6) In 32 examples showing positive expression of P21^WAF1/CIP1 , there were 20 examples showing positive expression of Survivin, the positive ratio was 62.50% (20/32),and in 28 examples showing negative expression of P21^WAF1/CIP1 , there are 27 examples showing positive expression of Survivin, the positive ratio was 96.43% (27/28), there was significant negative correlation difference between P21^WAF1/CIP1 and Survivin(r= -0. 411,P〈0. 005). Conclusion (1)The overexpression of CyclinD1, weaken or lost expression of P21^WAF1/CIP1 and high expression of Survivin may be associated with the generate and development of human astrocytoma. (2)And may indirect reflex the biological behaviour of human astrocytoma by detecting them in some extent.
